Spectral flow cytometry provides a set of challenges and opportunities for researchers. With the increased information that comes from measuring the full spectrum of light emitted by fluorescently tagged cells in flow, we resolve more markers than with conventional instruments. The increased complexity, though, requires extra precision in the construction of the assays, the panel design and the single-color controls that define the experiment. In this talk, I’ll try to provide some tips for getting the best out of your spectral flow cytometry experiments, drawing from recent advances in the field as well as some findings from our own lab on how to streamline your workflow and improve reproducibility.
