UCL Great Ormond Street Institute of Child Health Imaging Facility

The Imaging Facility provides light microscopy systems for researchers from UCL, Great Ormond Street Hospital, and the Institute of Child Health. We have three state of the art confocal systems for live and fixed cell imaging, two multiphoton system for deep tissue imaging, a live imaging widefield system, and an imaging suite for immunohistochemistry. Image processing and analysis includes Huygens deconvolution, Imaris 3D analysis and an in house 3 day ImageJ/Fiji course.

Expertise

Imaging Platforms

Keywords - Light Microscopy: Confocal microscopy | Widefield microscopy | Live cell imaging | Multi-photon microscopy

Zeiss LSM980 AiryScan2 Examiner Multiphoton confocal with Insight X3 multiphotonlaser
Zeiss LSM880 Examiner AiryScan Fast Multiphoton confocal with Mai Tai eHP DeepSee.
Zeiss LSM710 Observer confocal with 34 channel quasar detection.
Live imaging system (Nikon Ti2, Photometrix Kinetix camera, fully triggered high speed high sensitivity widefield)
Live imaging system (Zeiss Axiovert, hamamatsu Orca ER2, fully motorized stage, environmental control).
Spinning disk confocal (Ultraview CXU22) Widefield fluorescence and transmitted imaging suite with Zeiss, Leica and Olympus systems.

Applications

Keywords - Biological: Cell Biology | Neurobiology | Developmental Biology | Immunobiology | Zebrafish | Histology/pathology | ORGANOIDS | CLEARING

We specialise in live imaging of whole developing embryos, spheroids, and fixed and cleared tissue. In addition to our confocal systems we have a suite of microscopes for fluorescence, live cell imaging and histology which provide much needed basic imaging platforms that are heavily utilized as these systems are rarely found in a fully supported core facility.

Sample Preparation

Keywords - Biological: Resin embedding | Serial sectioning | Cryosectioning | Immunolabelling | Photoconversion | Immunofluorescence | Cryostat sectioning | Vibrating microtome | H&E staining | Live Cell

We have expertise in sample sectioning via cryostat or microtome. For whole embryo imaging we perform whole mount staining and use a variety of clearing techniques to image up to 5mm inside tissue.

Data Analysis

Keywords - Software: ImageJ | FIJI | Imaris | Matlab | Huygens | Volocity | AxioVision

We have 2 dedicated workstations with Huygens deconvolution, Imaris, Volocity, Matlab and various open source software. Training is offered for all software systems, and customized image analysis workflows can be provided.

Shared Access

The facility is open to all UCL staff and students. External users are welcome but should contact the facility manager for details.

Funding

Internally funded from UCL GOS Institute of Child Health and user access charges.
Supported by NIHR GOSH BRC.
